บทคัดย่อObjective : To evaluate the use and accuracy of the FACSCount^TM, Becton Dickinson, USA (BD) vs. the "gold standard" FACScan^TM (BD) method for determining absolute CD3, CD4 and CD8T-lymphocytes. Methods : From 3/94-9/94, 100 EDTA samples (30 controls and 70 HIV+/- tuberculosis inpatients) were analyzed on both the FACSCount and the FACScan using standard testing methods. For FACScan analysis, total WBC and % lymphocyte counts were determined by QBC^TM (BD) and FACScan 3-part differential, respectively. FACScan % and absolute cell counts obtained by SimulSET^TM software (BD) ; FACSCount absolute counts from the internal program. FACSCount CD4 counts reported as <50 were calculated as 25 and FACSCount CD8 counts reported as >2000 were calculated as 2000. Results : The correlation coefficients (R) for CD3, CD4 and CD8 absolute cell counts were all >= R=0.96, with very similar means/ranges. For both %CD4 and %CD8, and CD4/CD8 ratio,R=0.98. FACSCount requires less training and is less costly than standard FACScan.
